Judging from the old versions of the course (see: http://www.stanford.edu/class/cs221/index.html ) and the assignments & tutorials I would say/guess they are using Python for it. However it is very likely they will change/improve a lot of things in the course in order to enable it to scale up to the over 10.000 students that have signed up.
