incoming connections to which service? and this service is configured to listen on what ip/interface?

The private IP has not gateway assigned, so routes are not affected at all - I have checked & double checked this.

Is something changing the routes when you assign this interface the private IP?

Perhaps you should start by comparing the routes (netstat -rn) before
and after the
assignment of the private IP and see what paths are taken using traceroute.
